Quando il grande esploratore Dirk Pitt (Matthew McConaughey) trova una favolosa moneta collegata ad una leggenda storica, e inizia una caccia al tesoro attraverso alcune delle regioni più pericolose dell'Africa occidentale, si imbarca nell'avventura più importante della sua vita. Mentre cercano quello che la gente del posto chiama "la Nave della Morte" – una sperduta nave da guerra della Guerra Civile che protegge un carico segreto – Dirk ed il suo spiritoso compagno, Al Giordino (Steve Zahn), incontrano la dottoressa Eva Rojas (Penelope Cruz), una giovane brillante e bellissima che è convinta che il tesoro nascosto possa essere collegato ad un problema più ampio che minaccia il mondo che li circonda. A caccia di una nave della cui esistenza dubitano tutti gli altri, Dirk, Al, ed Eva devono affidarsi al loro ingegno e al loro coraggio per mettere nel sacco pericolosi signori della guerra, sopravvivere in terre minacciose, e riuscire a risolvere entrambi i misteri.
